Synthesis of Acetaminophen Chemistry of Medicine Luna Wang Introduction: Acetaminophen is a common drug used to reduce pain and fever. It is widely used to treat headaches, toothaches, muscle aches and menstrual period. It changes the way body senses pain and cools body. Overdoses of acetaminophen can cause severe liver damage. Acetaminophen was discovered in 1877 and introduced as a medicine by Heinrich Dreser in 1899. Objective: The purpose of this lab is to synthesize acetaminophen from a reaction between paminophenol and acetic anhydride and purify the product using recrystallization techniques. Reaction Equation: Figure 1: P-aminophenol reacts with acetic anhydride to form acetaminophen and acetic acid. Experimental: 1. Sand bath in a 500mL beaker was heated up to 120 Celsius by a hot plate. 2. About 0.41 g of solid p-aminophenol was weighted. 3. P-aminophenol was placed in a labeled glass test tube. 4. 1.2 mL of water and 0.5 mL of acetic anhydride was measured in a graduated cylinder and added to the test tube. 5. The solution was swirled to mix it. 6. The reaction mixture was dissolved in a 120 Celsius sand bath. 7. Once the solid material was dissolved, the reaction mixture was kept placing in the sand bath for an additional 20 minutes. 8. The test tube was removed from the heat after 20 minutes. 9. The reaction mixture was poured into a beaker and the test tube was rinsed with distilled water.
